Individual Human Venous EC Human Neutrophil Adherence


The chart above shows adherence of Human Peripheral Blood Neutrophils (PMN prepared by elutriation) to baseline and e.coli LPS-stimulated (100ng/ml 4 hrs, Sigma) confluent monolayers of 50 Individual (NON-POOLED) CSC 2VO V Human Venous Endothelial Cell (“HUVEC”) isolates: the frozen primary cell isolates were cultured and tested individually at 15 CPD (Passage 3) in the Complete Serum-Free Medium Kit With RocketFuel (SF-4Z0-500). Data is presented as simultaneous numeric adherence assessment of PMN to baseline (dark bar) and LPS stimulated (light bar) cell monolayers: values shown are arithmetic means of three replicate 24-well (2 cm2) cell cultures per individual donor per treatment. These data require (by Student’s T-test) that 6 individuals be tested in triplicate to achieve “P = < 0.05 significance” based on the 50-donor calculated parametric. Calculation of this statistic at the “P = < 0.01 significance” level requires that >12 individuals be tested. These cells represent the non-pooled analog of CSC 2VO V Human Venous Endothelial Cells. Pooled Human Venous EC Human Neutrophil Adherence


The chart above shows adherence of Human Peripheral Blood Neutrophils (PMN prepared by elutriation) to baseline and e.coli LPS-stimulated (100ng/ml 4 hrs, Sigma) confluent monolayers of 50 frozen aliquots of pooled CSC 2VO V Human Venous Endothelial Cells (“HUVEC”) pooled as primaries. Vials of pooled primary cells were thawed and tested in triplicate at 15 CPD (Passage 3) using the Complete Serum-Free Medium Kit With RocketFuel (SF-4Z0-500). Data is presented as simultaneous numeric adherence assessment of PMN to baseline (dark bar) and LPS stimulated (light bar) cell monolayers: values shown are arithmetic means of three replicate 24-well (2 cm2) cell cultures per pooled aliquot per treatment. These data require (by Student’s T-test) that 2 individuals be tested in triplicate to achieve the “P = < 0.01 significance” level. The cells evaluated in this test cells are CSC 2VO V.
